(A) Representative confocal microscopic images of colocalization of Beta 3 (β3) and filamin A in WT and migfilin-/- platelets spread on fibrinogen at the indicated time points. Experiments were repeated independently at least three times with similar results. Scale bars, 10 mm. (B) Colocalization of filamin A and β3 was quantified by Pearson Correlation Coefficient calculation using NIH Image J software. Results are expressed as mean ± standard error of the mean (SEM) from at least three independent experiments. Statistical significance was evaluated with the Student t test (*P<0.05, **P<0.01). (C) Washed WT platelets were stimulated with Mn2+ (0.5 mM) in the presence of fibrinogen (25 mg/mL) in suspension and lysed at indicated time points. Platelet lysates were immunoprecipitated with antibody against β3 and then immunoblotted with antibodies against filamin A and β3, respectively. Representative result from at least three independent experiments.
